Короткий огляд (реферат): Дисертаційна робота присвячена вивченню молекулярних механізмів взаємодії між альфа1-адреноблокаторами та адренорецепторами, сироватковим альбуміном людини, ДНК. У результаті досліджень розроблено QSAR-моделі залежності альфа1- адреноблокуючої активності похідних апорфіну та арилпіперазинів від квантово-хімічних та топологічних дескрипторів. Показано, що константи зв’язування арилпіперазинів з альфа1-адренорецепторами, отримані за допо- могою QSAR-моделі, статистично відповідають результатам досліджень на ізольованій сім’явиносній протоці щурів. Встановлено, що взаємодія сироваткового альбуміну людини з доксазозином супроводжується значною конформаційною перебудовою молекул альбуміну. Доксазозин у дослідженнях на культурах клітин HeLa та Hep G2 виявляє дозозалежний цитотоксичний ефект у концентраціях від 4 до 200 мкМ. Альфа1- адреноблокатор із групи арилпіперазинів має менш виражену властивість гальмувати проліферацію клітин, яка спостерігається при концентрації препарату 40 мкМ. Методом спектрофотометричного титрування та гель- електрофорезу підтверджено наявність фізико-хімічної взаємодії доксазозину з ДНК. Молекулярно-фармакологічний аналіз показує, що доксазозин зв’язується з ДНК по типу інтеркаляторів.
The thesis is devoted to the study of quantum-chemical and pharmacological properties of alpha1-adrenergic blockers to determine the quantitative structure-activity relationship (QSAR) in this group of drugs, as well as studying the molecular mechanisms of interaction between alpha1-adrenergic blockers and adrenoceptors, human serum albumin, DNA. The dependence of alpha1-adrenergic blockers (different chemical groups) pharmacological activity on the electronic and spatial properties of their molecules were determined. Physicochemical processes accompanying the binding of doxazosin and phenylephrine with human serum albumin, by 1H NMR spectroscopy were studied. Computer simulations of molecular mechanisms of interaction alpha1-adrenergic blockers with alpha1-adrenoceptors and human serum albumin by molecular docking have been performed. Research of the cytotoxic activity of alpha1-adrenergic blockers and analyzis the physicalchemical and quantum-chemical mechanisms of this process were made. As a result of research QSAR-models for predicting alpha1-adrenoceptor blocking activity of aporphine and arylpiperazine derivatives from quantumchemical and topological descriptors were developed. Affinity of aporphine derivatives to alpha1-adrenoceptors is determined by charges on carbon atoms C8, C11, C4 and topological descriptors (sum of valence degrees, topological diameter, the total molecular connectivity, Wiener index). Affinity of arylpiperazine derivatives to alpha1-adrenoceptors is determined by charges on nitrogen N10, carbon C12, C6, C8 atoms, electrostatic potential of carbon atoms C1, C5, binding constant to albumin, and dipole moment, molecular weight, topological diameter of molecules. Doxazosin and arylpiperazine derivative displayed similar potency in vas deferens (pA2=8.51±0.8 and 8.85±0.9, respectively). It has been shown that thebinding constants for arylpiperazines with alpha1-adrenoceptors predicted by QSAR-model are statistically consistent with the results of studies on isolated rats vas deferens. The interaction of human serum albumin with doxazosin and phenylephrine at a concentration of 0.1% of drugs causes a decrease in the volume of all types of water-filled compartments in the protein globule and is accompanied by a significant conformational rearrangement of albumin molecules. The results of molecular docking suggest that binding center of adrenoblokers includes amino acids in alpha1-adrenergic receptor: Ser192, Asp106, Ser188, Gln177, Ile178, Tyr316, Met292; in human serum albumin: Leu115, Met123, Phe134, Phe149, Tyr161, Leu185, Arg186, Leu185, Ile142. Human serum albumin and alpha1-adrenergic receptor complexes with adrenergic blockers are stabilized by hydrogen bonds and hydrophobic interactions. Doxazosin in cytological studies on cultures of HeLa and Hep G2 cells exhibits a dose-dependent cytotoxic effect at concentrations ranging from 4 to 200 μM. Alpha1-adrenergic blocker from the arylpiperazines group has a less pronounced ability to inhibition of cell proliferation, which is observed at drug concentrations 40 μM. Spectral changes in UV absorption titration shows the interaction of doxazosin with the DNA double helix. The binding constant for the interaction between doxazosin and DNA was K = 99 (±0,6) M-1 and the binding site size was 1,7 (±0.3) base pairs. Destruction of DNA with increasing concentrations of the doxazosin in agarose gel has been observed. Molecular pharmacologic analysis showed that doxazosin is associated with the DNA as intercalator. The dependence of the cytotoxic effect of alpha1-adrenergic blockers from chemical structure is a theoretical basis for synthesis of new compounds that do not affect cell proliferation and show no adverse effects on the cardiovascular system. Data about doxazosin binding site in human serum albumin expands knowledge of this drug pharmacokinetics and enable prediction of its interaction with other drugs, characterized by high affinity to plasma proteins. The results are methodologically important for pharmacology and pharmacy. Proposed methods of quantum-chemical and topological descriptors calculation for the receptortargeted dugs, and their statistical analysis can be used to build predictive QSARmodels for compounds of other pharmacological groups.
