Краткий осмотр (реферат): Проведено експериментальне дослідження антигіпоксичних властивостей похідних глюкозаміну на моделях гіпобаричної гіпоксії у мишей, гемічної гіпоксії та гіперволемічного набряку головного мозку у щурів. Виявлено антигіпоксичні, антиоксидантні властивості похідних глюкозаміну в умовах церебральної гіпоксії та обгрунтовано вибір найбільш перспективного аміноцукру для подальших досліджень – глюкозаміну гідрохлориду. Досліджено антиамнестичну активність глюкозаміну гідрохлориду та виявлено умовно ефективну дозу – 50 мг/кг. При вивченні фармакологічних властивостей глюкозаміну гідрохлориду при гострому порушенні мозкового кровообігу (у гострому та відновлювальному періодах) встановлено, що досліджувана речовина збільшує виживаність, знижує ступінь неврологічного дефіциту та покращує рухову активність щурів. Також виявлено властивості глюкозаміну гідрохлориду стабілізувати енергетичний метаболізм, гальмувати процеси вільнорадикального окиснення та стимулювати антиоксидантну систему головного мозку щурів. Застосування глюкозаміну гідрохлориду супруводжувалось збереженням морфологічної структури головного мозку, її кількісного та якісного складу, функціональних властивостей, що підтверджує його церебропротекторну дію. Імовірний механізм церебропротекторної дії ГА г/х полягає у гальмуванні процесів утворення АФК та стабілізації біологічних мембран. Одним з важливих аспектів церебропротекторної дії глюкозаміну гідрохлориду є гальмування апоптозу, що доведено дослідженнями in vitro та in vivo.
The dissertation work is devoted to the experimental investigation of the cerebroprotective activity of glucosamine and its derivatives. The antihypoxic activity of glucosamine derivatives was investigated using the models of hypobaric hypoxia in mouses, hemic hypoxia and hypervolemic brain oedema in rats. On all three models the use of glucosamine derivatives was associated with the prolongation of the animal’s life time, decrease of the brain mass coeficient and changes of N-acetylglucosamine methabolism in brain and in liver. Glusosamine derivatives have shown antihypoxic and antioxidant activity in the terms of experimental hypoxia. Changes of the N-acetylglucosamine methabolism enable to conclude that it is supporting the antihypoxic activity of glucosamine derivatives. On tha basis of the screening inversigations was done the choice of the most perspective aminosugar for further studies – glucosamine hydrochloridum. The antiamnestic activity of the glucosamine hydrochloridum was investigated using the conditional reflex of passive avoidance for the assessment of the cognitive and mnestic functions. Glucosamine hydrochloridum was given in dosages 50, 100 and 200 mg/kg and 24 hours after amnesia was revealed. The increase of dose was not associated with efficieany improvement, the relative effective dose − 50 mg/kg was detected. Further investigation of the pharmacological properties of glucosamine hydrochloridum was conducted using the model of acute damage of the cerebral blood flow (in acute and in early rehabilitation period). It was determined, that the inverstigated substance increases the survival rate, that was estimated on the level of 70% in the acute period (4th day) and 65% in the early rehabilitaion period (18th day) and was comparable with reference medicine Mexidolum. The administration of glucosamine hydrochloridum was associated with decrease of the level of neurological deficiency (McGrow skale), and increase of the motion activity of rats that on 18th day after acute damage of cerebral blood flow was comparable with the level of healthy rats. The properties of glucosamine hydrochloride to improve the energy and carbohydrate methabolism in the rat’s brain tissue were determined during the study. The adenylic nucleotides sum and phosphorylation index by glucosamine hydrochloridum administration achieved the level of healthy rats on the 18th day of ishemia, meanwhile the lactate level was ca. 40% lower than the control group level. The properties of glucosamine hydrochloridum to inhibit the processes of free-radicals oxidation were detected. The concentration of the primary and secondary products of lipids peroxidation in the glucosamine hydrochloridum group were significantly lower compairing to the control group and reference medicine. On the 18th day of ishemia the TBA-reactants concentrations in the brain had no significant difference in groups of glucosamine hydrochloridum and healthy rats. The administration of the studied substance is also associated with significant lower levels of oxidative proteins modification markers, that at the end of early rehabilitation period decreased to the level of healthy animals. The activation of iNO-synthase that is specific for the ishemic injuries of brain was by glucosamine hydrochloridum use significant lower, that resulted in lower levels of NO-intermediates. The glucosamine hydrochloridum administration was associated also with stimulation of the antioxidant system that resulted in significant higer levels of cyclooxygenase and catalase compairing to the control group and mexidolum group. The morphological investigations showed that glucosamine hydrochloride was saving the morphological structure of the brain, its qualitative and quantitative composition, its functional potential, confirming its cerebroprotective effect. The probable mechanisms of the glucosamine hydrochloridum cerebroprotective activity is based on the inhibition of the active oxigen forms formation and stabilization of the biological membranes. To one of the important aspects of cerebroprotective activity of glucosamine hydrochloridum belongs inhibition of apoptosis, that was proven by investigations in vitro and in vivo.
