Теоретичне обґрунтування та розробка комбінованих лікарських препаратів для лікування мастопатії на основі лікарської рослинної сировини

Abstract

Дисертація присвячена науковому обґрунтуванню складу та розробки про-мислової технології та стандартизації нових оригінальних лікарських препаратів: бігелю комплексної дії «Мастонорм», мазі на основі фітоолій «Фітолан», багато-компонентного збору «Маммофіт», для комплексної фармакокорекції мастопатії. Із використанням сучасних методів дослідження вивчено фізико-хімічні вла-стивості розроблених препаратів, запропоновано методики якісного та кількісного аналізу діючих речовин у них, розроблено проєкти МКЯ на оригінальні ЛЗ – бі-гель комплексної дії «Мастонорм», мазь на основі фітоолій «Фітолан» та багато-компонентний збір «Маммофіт», екстемпоральна суспензія. Проведено валідацію технологічного процесу та аналітичних методик розроблених препаратів.

Диссертация посвящена научному обоснованию состава и разработки промышленной технологии и стандартизации новых оригинальных лекарственных препаратов: бигеля комплексного действия «Мастонорм», мази на основе фитомасел «Фитолан», многокомпонентного сбора «Маммофит», для комплексной фармакокоррекции мастопати. С использованием современных методов исследования изучены физико-химические свойства разработанных препаратов, предложены методики качественного и количественного анализа действующих веществ в них, разработаны проекты МКЯ на оригинальные лекарственные средства: бигель комплексного действия «Мастонорм», мазь на основе фитомасел «Фитолан» и многокомпонентный сбор «Маммофит», экстемпоральная суспензия. Проведена валидация технологического процесса и аналитических методик разработанных препаратов.

The thesis is devoted to the scientific substantiation of the composition and development of industrial technology and standardization of new original medicinal preparations– complex action bigel «Mastonorm», ointments based on phytomases «Fitolan», multicomponent collection «Mammofit» for complex pharmacological correction of mastopathy. With the use of modern research methods, the physicochemical properties of the developed drugs were studied, methods of qualitative and qualitative analysis of the active substances in them were proposed, MLC projects were developed for original drugs– Mastonorm complex action bigel, ointment based on phytomases «Fitolan» and multicomponent collection «Mammofit». For the first time the methodological approach to pharmaceutical development of combined medicines on the basis of medicinal plant raw materials for application in the scheme of complex therapy of mastopathy was substantiated. Data from the scientific literature on the etiopathogenesis, diagnosis and modern methods of pharmacotherapy of mastopathy are analyzed. Based on the study of the range of drugs for the treatment of pathology in Ukraine, the expediency of expanding the range of drugs for the comprehensive treatment of mastopathy, including on the basis of raw materials of natural origin was substantiated. Based on the results of marketing research and analysis of scientific literature, the feasibility of bringing to the domestic pharmaceutical market of original phytopreparations for complex pharmacocorrection of mastopathy was substantiated. To solve the tasks, the work used organoleptic (appearance, odor, color), pharmacological (moisture content, fractional composition of MPRM, fluidity, bulk and bulk density, dispersion of emulsion and suspension systems, colloidal and thermal stability, structural and mechanical properties, texture analysis, etc.), physicochemical (solubility, relative density, pH, dry residue, UV spectrophotometry, thin layer chromatography, etc.), biopharmaceutical (release of biologically active substances from soft DF «in vitro»), microbiological (effectiveness of antimi» A general algorithm for the optimal composition of the bigel has been developed under the conditional name «Mastonorm»: based on the results of a set of physicochemical, pharmacotechnological, biopharmaceutical studies, the choice of gel formers of the hydrophilic and hydrophobic fraction of the bigel was substantiated; according to the results of physico-chemical and biopharmaceutical studies, ethanol was introduced into the gel at a concentration of 6 %; the need to use preservatives to ensure the shelf life– nipagin– 0.4 %, nipazol 0.1%; rheological and thermogravimetric studies substantiate the phase ratio in the composition of the bigel «Mastonorm»; using the methods of mathematical planning of the experiment, the substantiation of the optimal composition of the bigel was carried out. The general algorithm of definition of optimum structure of ointment under the conditional name «Phytolan» was developed:– by results of bibliosemantic researches the choice of phytooils was substantiated; according to the results of a complex of physico-chemical, pharmacotechnological microbiological researches the composition of ointment on the basis of phytooles under the conditional name «Fitolan» was established; the spectrum and level of antimicrobial activity of the drug were studied; developed methods of API standardization and studied the stability of the ointment during storage. The technological process and analytical methods of the developed preparations were validated.
