Теоретичне та наукове обґрунтування фармацевтичної розробки комбінованих лікарських препаратів для комплексного лікування гельмінтозів системи травлення

Abstract

Дисертаційна праця присвячена комплексним дослідженням щодо тео-ретичного та експериментального обґрунтування створення ЛП комбінованого складу для комплексного лікування гельмінтозів системи травлення у дорослих та дітей. Уперше обґрунтовано методологічний підхід до створення ЛП для фармакотерапії гельмінтозів травної системи на різних етапах лікування, на під-ставі якого проведено фармацевтичну розробку таблеток, вкритих оболон-кою, пастилок жувальних та капсул. Обґрунтовано склад, раціональну технологію та методи стандартизації ЛП «АП-гельмін (на основі альбендазолу та празиквантелу), «Альбенпаст» (на основі альбендазолу), «Фітогельмін» (на основі сухих екстрактів ЛРС). Розроблено та запропоновано лінійку ЛП із протигельмінтною активністю для екстемпорального виробництва. Визначено види первинного упаковання, встановлено умови зберігання і терміни придатності розроблених ЛП. Розроблено проєкти технологічного регламенту та МКЯ на створені ЛП. Доклінічними дослідженнями підтверджено широкий спектр специфічної активності та низький ступінь їх токсич-ності.

Диссертация посвящена комплексным исследованиям теоретического и экспериментального обоснования создания лекарственных препаратов комбинированного состава для комплексного лечения гельминтозов системы пищеварения у взрослых и детей. Впервые обоснован методологический подход к созданию ЛП для фармакотерапии гельминтозов пищеварительной системы на различных этапах ле-чения, на основании которого проведено фармацевтическая разработка таблеток, покрытых оболочкой, пастилок жевательных и капсул. Обоснованы состав, рациональная технология и методы стандартиза-ции ЛП «АП-гельмин» (на основе альбендазола и празиквантела), «Альбен-паст» (на основе альбендазола), «Фитогельмин» (на основе сухих экстрактов ЛРС). Разработана и предложена линейка ЛП с противогельминтной активно-стью для экстемпорального производства. Определены виды первичной упаковки, установлены условия хранения и сроки годности разработанных ЛП. Разработаны проекты технологического регламента и МКЯ на созданные ЛП. Доклиническими исследованиями подтверждены широкий спектр специфической активности и низкая степень их токсичности.

The dissertation is devoted to complex studies of the theoretical and experi-mental substantiation of the creation of drugs with a combined composition for the complex treatment of helminthiasis of the digestive system in adults and children. For the first time, a methodological approach to the creation of drugs for the pharmacotherapy of helminthiasis of the digestive system at various stages of treat-ment was substantiated, based on which the pharmaceutical development of coated tablets, chewable pastilles and capsules was carried out. For the first time, on the basis of a number of studies, the composition and technology of drugs “AP-helmin” (based on albendazole and praziquantel), “Albenpast” (based on albendazole), “Phytohelmin” (based on dry extracts of medicinal products) were theoretically substantiated and experimentally developed. For the first time, it was theoretically substantiated and proved by experimental studies of the specific pharmacological activity of the expediency of combining albendazole and praziquantel in the tablet form. A study of the physicochem-ical and pharmacological properties of a mixture of APIs and tablets based on them was carried out. For the first time, a children's dosage form for the treatment of helminthiasis of the digestive system based on albendazole was proposed – chewing pastilles. The composition of auxiliary substances, manufacturing technologies have been processed and critical production parameters have been established. For the first time, a drug in the form of capsules based on a complex of dry medicinal plant extracts was proposed for the 3rd stage of treatment of helminthia-sis of the digestive system (elimination of the consequences of the disease and res-toration of the body). The choice of API and excipients is substantiated. Three original new drugs, “AP-helmin”, “Albenpast”, “Phytohelmin” for the treatment of helminthiasis of the digestive system were developed and proposed for practical pharmacy and medicine. Regulatory and technological documentation for the original drugs “AP-helmin”, “Albenpast”, “Phytohelmin” was developed. Preclinical studies of the drugs were carried out, which confirmed the presence of specific activity and a low degree of their toxicity. A line of drugs with anthelmintic activity for extemporal production was developed and proposed. Methodological recommendations for the creation of drugs with anthelmintic activity based on medicinal plant raw materials and their derivatives “Pharma-cotherapy of helminthiasis of the digestive system and the development of anthelmintic phytopreparations: analysis of domestic and world experience” was developed and approved by the Scientific Council of the NUPh. Some fragments of the work were introduced into the research work and educational process of a number of pharmaceutical and medical institutions of higher education in Ukraine.
